a Schematic representation of the cell culture eGFP silencing experiment. Cells are incubated with LNPs for 24 hours, followed by medium replacement and culturing for an additional 48 hours. At 72 hours, the cells are collected and analyzed using FACS for LNP uptake and eGFP silencing. b, c LNP uptake and eGFP silencing in U2OS-UBE21-eGFP or HeLa-CD63-eGFP cell lines. LNP uptake is quantified based on the mean fluorescence intensity (MFI) of DiD in the cells. The relative eGFP fluorescence (%) is quantified as the MFI of GFP in the treated cells compared to PBS treated cells.
