Oxygen consumption rate (OCR) was determined throughout the mitochondrial respiration test in control and patient derived P525L fibroblasts. Arrows indicate the time when mitochondrial inhibitors were added to the media to assess respiratory parameters. Non-mitochondrial oxygen consumption was determined by measuring the difference between total oxygen consumption and antimycin A and rotenone treatment induced reduction in oxygen consumption, maximal respiration was expressed as difference between oxygen consumption following mitochondria uncoupling by FCCP and rotenone, antimycin A treatment and spare respiratory capacity was determined by subtracting basal respiration from maximal respiration. The data represent mean ± s.e.m. from three independent experiments and eight individual points are plotted in the graph. All statistical analysis were performed by two-sided student-t test using graph pad prism software. Source data are provided as a Source Data file.
