Mycelia of the parental and pTetOff-erg6 strains were grown in GMM broth without doxycycline (DOX) (A) and with 4 μg/ml doxycycline (B) for 16 h at 30 °C. Hyphae were subsequently stained with 25 μg/ml filipin for 5 min. Fluorescent images were captured using DAPI filter settings. White arrows indicate sterol-rich plasma membrane domains (SRDs). Scale bar = 10 µm. Microscopy experiments were completed three times independently with similar results.
