Fig. 4: Structurally distinct melanins from diverse fungal sources block airway epithelial CXCL8.
From: Fungal melanin suppresses airway epithelial chemokine secretion through blockade of calcium fluxing

a H292 cells were stimulated for 6 h with wildtype A. fumigatus (B5233; 1 × 107/cm2), ΔpksP conidia (1 × 107/cm2) with or without C. neoformans H99 melanin ghosts (Cn-MG) (5 × 107/cm2), P. aeruginosa PAO1 strain (PSA) with or without Cn-MG, or Cn-MG alone. CXCL8 apical secretion was measured by ELISA. See also Figure S4. b H292 epithelium was infected with PSA in the absence or presence of synthetic L-DOPA-coated FLP (5 × 107/cm2), β−1,3-glucan-coated FLP (BG; 5 × 107/cm2), or naked FLP (5 × 107/cm2) for 6 h, then apical CXCL8 secretion was measured by ELISA. Media or coated beads alone were included as negative controls. Data are represented as mean ± SEM. n = 3 from three replicates; One-Way ANOVA with Dunnett’s multiple comparisons test; *p < 0.049, **p = 0.0004, ****p < 0.0001 compared to stimulation alone (i.e., ΔpksP or PSA alone).
