Gene transcription is controlled by elements such as promoters, cis-Regulatory Elements (cREs), and anchors within Insulated Genomic Domains. A systematic approach based on a library of Epigenomic Controllers (ECs) allows precision control of gene programs through modulation of epigenetic state. Briefly, epigenomic effectors, chosen from an in-house library, are fused to customized DNA binding domains to form ECs. These ECs target specific elements that control transcription and can be applied to induce a tuned transcriptional response of one or multiple Insulated Genomic Domain (IGD) genes.
