a, b RP-HPLC traces (214 nm, 60 min 0–80% MeCN gradient) and corresponding ESI-MS of the lysine fatty acid functionalized Semaglutide (4) and the all-hydrocarbon stapled NYAD-13 (5). All peptides were synthesized in full automation, including assembly, chemical modification, cleavage, and ether workup, and were purified by RP-HPLC. Mass spectra were obtained from the summation of the entire HPLC peak.
