a-g, Localizations of Venus-tagged proteins in various Chlamydomonas mutant backgrounds. Transparent arrowheads point to membranes inside the matrix; filled arrowheads point to membranes at the matrix entry sites. Images are presented at the same scale. h-j, Two-color immunofluorescence was performed for anti-FLAG and anti-CAH3 (h,i) and anti-FLAG and anti-MITH1 (j). Images are presented at the same scale. k, Immunofluorescence controls showing background signal in the region of the pyrenoid. Images are presented at the same scale as in (h). Control images are thresholded such that their intensity ceiling is matched to the experiment(s) which used that combination of antibodies. For the cah3 control, the FLAG and CAH3 channels were thresholded so that their upper intensity limit matched that of the FLAG and CAH3 images showing the weakest signal across Fig. 3h, i and Extended Data Fig. 5h, i. For the mith1 control, the FLAG and MITH1 channels were thresholded so that their upper intensity limit matched that of the FLAG and MITH1 images showing the weakest signal across Fig. 3j and Extended Data Fig. 5j.
