Analysis of fetal mammary rudiments heterozygous (Lgr5KIHET) and homozygous (Lgr5KIHOM) for the Lgr5KI allele. a Fetal mammary rudiments were isolated from Lgr5KIHET and Lgr5KIHOM littermates at stage E18 and stained in whole mount for GFP (top, bar represents 200 μM), and Keratin 8 (K8, green) and Keratin 14 (K14, red, bottom). b Single-cell suspensions were prepared from Lgr5KIHET and Lgr5KIHOM littermates at stage E18. Live, single, epithelial (EpCAMPOS) cells were analyzed for GFP expression. c Single-cell suspensions were prepared from Lgr5KIHET and Lgr5KIHOM littermates at stage E18. Live, single, epithelial cells were analyzed for EpCAM and CD49f expression by FACS. d Single-cell suspensions were prepared from Lgr5KIHET and Lgr5KIHOM littermates at stage E18. Live, single, EpCAMHIGH; CD49fHIGH; GFPPOS cells were sorted and plated in to 3D culture containing 2% Matrigel.
