Error bars represent mean ± SD. Results are from three independent experiments. Unpaired two-tailed Student’s t-test with 95% CI was used to determine difference, p < 0.05 is considered significant. c,d, ETD-MS/MS spectra of two O-GlcNAcylated peptides of recombinant GFP-SynGAP CC-PBM purified from HEK293T co-expressing OGT. The c2 ion unambiguously confirms O-GlcNAcylation at S1159 (c). The c12 and c14 ions unambiguously confirms O-GlcNAcylation at T1306 (d). The matched fragment ions are marked.
