a. C to T damage profiles generated in Damage Profiler show characteristic peaks at fragment ends, with an uptick beyond ~20 bp reflecting damage from the opposite end of the short molecules. b. Base composition around fragment termini, generated in mapDamage, reveals guanine and adenine enrichment immediately upstream of reads (gray box), consistent with depurination driven strand breaks in ancient DNA. c. Read length distribution plot (DamageProfiler), with a minimum length threshold of 30 bp applied.
