a, HeLa cells were stably transduced with lentivirus containing tetracycline-inducible SLFN5-HA genes (WT and Δ730-891). SLFN5-HA was induced with doxycycline (0.5 μg/ml) for 48 h. SLFN5 expression was confirmed by immunoblot analysis. b, c, Cells were infected with HSV-1 (b) and ΔRING virus (c) at an MOI of 0.1. At 24 hpi, viral DNA was measured by qPCR. n = 3 biologically independent experiments. Data are the mean ± SD. Comparisons between groups were performed using the two-tailed unpaired Student’s t-test. **, p < 0.005. n.s., not significant.
