High-fat diet depletes skin Treg cells

Human obesity is associated with inflammatory skin conditions such as psoriasis, and high-fat diet (HFD)-induced obesity in mice exacerbates psoriasis through the accumulation of IL-17-producing γδ T cells. Sivasami et al. identify a population of PPARγ+ regulatory T (Treg) cells in skin that normally restrain inflammation but are depleted by HFD.

RNA-sequencing (RNA-seq) showed that Pparg transcripts are highly enriched in skin Treg cells compared with spleen Treg cells. A large fraction of Treg cells in trunk and ear skin of mice were PPARγ+, mainly found in the dermis near to γδ T cells, and these were more proliferative and produced more IL-10 than PPARγ– skin Treg cells. Published single-cell RNA-seq datasets of mouse and human skin Treg cells similarly showed the presence of a PPARγ+ subset. Treg cell-specific ablation of PPARγ reduced the size of the Treg cell compartment in skin, but not in spleen, and led to dysregulation of the unique gene expression signature of skin Treg cells at steady state.
