a, A simplified schematic of REAP. Antibodies are incubated with a barcoded yeast library displaying members of the exoproteome. Antibody-bound yeast are enriched by magnetic-column-based sorting and enrichment is quantified by next-generation sequencing. b, Heat map of all profiled reactivities across all patient samples stratified by disease severity and using the same colour scale as in Fig. 1a. Sticky antigens (as defined in Methods) were removed from the heat map. c, SARS-CoV-2 RBD REAP scores for samples from patients with COVID-19 stratified by positive or negative ELISA RBD reactivity. d, IL-6R REAP scores for samples from patients with COVID-19 stratified by treatment with an anti-IL-6R biologic therapy (tocilizumab or sarilumab). Samples collected at least one day after infusion were considered treated. Samples collected on the day of infusion were excluded from analysis owing to uncertainty in the timing of sample collection. Significance in c, d was determined using a linear mixed model (Methods). In c, d, n values include longitudinal samples from the same patient.
