N cleavage was detected with Western blots. c 293T cells were transfected with an IFN-β-Luc reporter plasmid, expression constructs of caspase-6 and coronavirus N, with or without poly(I:C). Cells were incubated for 24 h before harvesting for dual-luciferase reporter assays (n = 3). d 293T cells were transfected with the same set of plasmids. Gene expression of IFIT3 and OAS1 was quantified with RT-qPCR (n = 4). Experiments in (a, b) was repeated three times independently with similar results. Data represented mean and standard deviations from the indicated number of biological repeats. Statistical significance between groups was determined with one way-ANOVA (c, d).
