ROCK2 (rho-associated coiled-coil-containing protein kinase-2) is a protein with multiple functions, and its activity is upregulated in acute inflammatory injury and chronic diseases such as diabetes, metabolic syndrome and idiopathic pulmonary fibrosis. Blocking ROCK2 acts by quelling pro-inflammatory TH17 cells and boosting regulatory T cells to restore homeostasis. In September the FDA approved Jakafi (ruxolitinib) for chronic GVHD after failure of one or two lines of systemic therapy.
