Supplementary Figure 7

(a) Confocal images of the spinal cord of iNOS-tdTomato-cre x Arginase-YFP mice perfused at EAE onset (tdTomato, red; YFP, green) illustrating spatial distribution of the polarized cells within the lesion based either on 20µm–thick bands starting from pia surface (left panel), concentric squares starting from lesion center (middle panel), or bands orientated in parallel to a central blood vessel (laminin in cyan; right panel). Scale bar 20 µm. (b) Quantitative analysis of the proportion of MiNOS, MiNOS/Arginase and MArginase cells in lesions subdivided as indicated in (a). Shown is average ± s.e.m.; n = 7 mice and 1,869 cells assessed for the analysis starting from the pial surface (left panel), n = 7 mice and 1,894 cells assessed for the analysis from the center of the lesion (middle panel), n = 5 mice and 436 cells assessed for the analysis starting from the blood vessel (right panel). No significant differences were observed (2-way-ANOVA with Bonferroni post-hoc correction).
