Dynamic modifications of RNAs play essential regulatory roles during RNA-processing events and thereby affect gene expression as well as cellular functions, development and diseases such as cancer. Dan Dominissini and Gideon Rechavi illustrate the broad relevance of ‘epitranscriptome regulation’, as this phenomenon is now known, using as an example the functions and mechanisms of action of N 6-methyladenosine (m6A), an abundant and well-studied RNA modification.
