Optimization of the JF646-HTL concentration for visualization of single-molecules of Hht1-HaloTag. Log phase cells of MP792 were treated for 30 min with four different concentrations of JF646-HTL (0 nM, 1 nM, 5 nM, 10 nM). Cells were washed twice with EMM media to remove excess dye and imaged using HILO illumination mode. Cells were classified according to the labeling density: No labeling (orange arrowheads), single-molecule labeling (blue arrowheads), and overcrowding (pink arrowheads).
