Effects of co-expression of the BirA gene and biotin bait on sRAGE biotinylation. (A) Generation of transgenic silkworms expressing both the sRAGE and BirA genes. Adult transgenic silkworms expressing the sRAGE gene or BirA gene were mated. During the fifth instar, approximately 20 μg/g biotin bait was fed to the larvae. The MSGs were isolated from the F1 larvae, and total proteins were extracted. (B) CBB staining of SDS-PAGE gels. Proteins extracted from MSGs were separated by SDS-PAGE followed by staining with CBB. (C) Detection with streptavidin-HRP. Lysates were separated by SDS-PAGE, followed by western blotting with streptavidin-HRP. Protein extracted from MSGs of transgenic silkworms harboring only the Ser1-GAL4 construct was used as a negative control (lane 1). Numbers at the left indicate molecular masses (kDa).
