Algorithm optimization. (a) Linear fit of the mean operator counts versus the numbers of microglia detected by the automated algorithm, reveals a strong correlation (y = 1.007x; R2 = 0.843) and validates our algorithm. (b) This is confirmed by the Bland-Altman plot, which shows a bias of 4.20%. For comparison, bias for three observers was −8.35%, 4.33% and 3.50%.
