Functional non-redundancy of YAP and TAZ revealed by cell-based luciferase assay. (A) YAP knockdown cells show higher reporter activity than TAZ knockdown cells, in the presence of a synthetic 8xGTIIC TEAD luciferase promoter. YAP or TAZ knock down efficiency was checked with antibodies and GAPDH was used as loading control. (B) In the presence of a single TEAD binding site at the enhancer, control, YAP and TAZ transfected cells showed similar reporter activity. In the presence of tandem TEAD binding sites, YAP transfected cells showed double the reporter activity (additive), while TAZ transfected cells showed more than double the reporter activity. Each bar represents the mean and standard deviation of three independent experiments. (C) Proposed model of how the formation of a TAZ-TEAD heterotetramer complex may result in a stronger additive effect in the presence of tandem TEAD binding sites (blue block arrows).
