Immunofluorescence staining of EF-Tu on bacterial cell surface of EUU vs. EUK. Bacterial cells were fixed with 4% paraformaldehyde (without permeabilization as to stain only the surface EF-Tu, not its cytoplasmic form) and then incubated with mouse monoclonal anti-EF-Tu antibody followed by incubation with Alexa Fluor 555-conjugated donkey anti-mouse secondary antibody containing 0.1 μg/ml Hoechst dye for DNA staining. The immunofluorescence micrographs were then obtained using ECLIPSE Ti-Clsi4 Laser Unit (Nikon) equipped with NIS-Elements D V.4.11 (Nikon). Original magnification power was 630X for all panels. Expression of surface EF-Tu is shown in red, whereas DNA contents are illustrated in blue. DIC = differential interference contrast mode of image capture.
