Effects of oxygen tension, BMP-2 and/or TGF-β1 on the mRNA levels of nonspecific cartilage markers and the protease HtrA1. hBM-MSCs were cultured in type I collagen sponges for 3, 7, 14, and 21 days as described in Fig. 3. The levels of mRNA encoding type I collagen (COL1A1 mRNA) (A), type X collagen (COL10A1 mRNA) (B) and HtrA1 (HtrA1 mRNA) (C) were measured using real time RT-PCR with specific primers. All the results were normalized to RPL13a mRNA and are presented as the expression of each gene relative to that of undifferentiated hBM-MSCs at day 0. Box plots represent independent experiments performed in triplicate (n = 4). Statistically significant differences between the undifferentiated cells at day 0 and treated cells are presented and were determined using the Kruskall-Wallis test (*P < 0.05).
