(A) TRAP expression in the synovium of mice with CIA (mock or Ssu72-overexpressing) was observed using immunohistochemical staining (original magnification, 200× or 400×, n = 6). (B and C) Bone marrow cells from mice with CIA (mock or Ssu72-overexpressing) were cultured with macrophage colony-stimulating factor (M-CSF) (10 ng/ml) and RANKL (50 ng/ml). Cells were fixed, stained for TRAP, and the number of TRAP+ cells was counted using a light microscope (original magnification 100×, n = 6). Real-time PCR was performed to measure the relative mRNA levels of osteoclastogenic markers (n = 6). The data represent the mean ± SD from three independent experiments. Statistical analyses were conducted using the nonparametric Mann-Whitney U-test (*P < 0.05, **P < 0.03, ***P < 0.01).
