Radiation effectively induces T-cell inactivation and upregulates NKG2D ligands and CD48 in human PBMCs. The proportion of T cells and expression of NKG2D ligands and CD48 were measured by flow cytometry (FCM). (A) Results of phenotypic analysis by two-color FCM after 21 days. (B) Ratios of mean fluorescence intensities (MFIs) obtained from irradiated PBMCs and untreated control (fresh PBMCs). Relative expression ratios were calculated by dividing irradiated PBMC MFI by fresh PBMC MFI. (C) Representative FCM histograms (white: isotype control, gray: fresh PBMCs, black: 72 h after irradiation). All the experiments were conducted in triplicate for each donor (n = 5). The statistical significance was determined using paired Student’s t test. *P < 0.05, **P < 0.005, ***P < 0.0005 (*fresh PBMCs versus irradiated PBMCs).
