Increased adenosine induced by high D-glucose is restored by insulin in rat glomeruli. (A) Purified glomeruli from healthy male rats were exposed ex vivo to 5mM or 25mM D-glucose (D-glc) and insulin 10 nM for 24 h. Extracellular adenosine was quantified in aliquots of the medium by using derivatization with chloroacetaldehyde and HPLC. Values are means ± S.D. from individuals determinations normalized to 1 µg of total glomerular proteins. *P < 0.01 vs 5mM; #P < 0.01 vs 25mM D-Glc; n = 6. (B) Time course of extracellular adenosine levels in glomeruli of rats exposed ex vivo to 25mM D-glucose and supplementation with insulin at 0.2, 0.5, 2 and 10nM at 24 h.
