Total protein sulfenylation and SIRT6 specific sulfenylation is increased in THP-1 cells and SIRT6 sulfenylation is increased in human monocytes stimulated with LPS and in mouse spleen subjected to CLP-induced sepsis: (A) Effect of LPS stimulation on sulfenylation in THP-1 cells: Cells were stimulated with LPS over a time course of 0–24 hours and lysates were evaluated for total protein sulfenic acid formation (n = 3), (B) Mass spectrometry analysis identified sulfenylated proteins at 3 and 6 h time points mapping to SIRT6 interactome. (C) Evidence of SIRT6 sulfenylation: Cells were stimulated with LPS over a time course of 0–24 hours and lysates were evaluated for SIRT6 sulfenylation (n = 5). (D) Human adherent monocytes isolated from peripheral blood were stimulated with LPS over a time course of 0–24 hours and lysates were evaluated for SIRT6 sulfenylation (n = 3).
