UVB-induced oxidative skin damage was attenuated in fat-1 transgenic and DHA-treated WT mice. (A,B) Isolated protein extracts were electrophoresed and transferred to a PVDF membrane, which was probed using antibody for 4-HNE. Results are expressed as means ± SE (n = 5 per treatment group). ***p < 0.001 versus UVB-irradiated WT mice; *p < 0.05 versus UVB-irradiated control group. Formalin-fixed and paraffin-embedded tissues from UVB-irradiated mice were immunostained for detecting the 4-HNE (upper panels)- or MDA (bottom panels)-modified protein expression. Magnifications, ×100 (4-HNE) and ×200 (MDA). Scale bar is 100 µm. (C) Animal treatment and other experimental details are same as described in the legend to Fig. 3. The respective sections were immunostained for 4-HNE and MDA, and counterstained with hematoxylin. Positive 4-HNE and MDA staining yielded a brown-colored product. Magnifications, ×200.
