Validation and optimization of Exchange-PAINT for membrane receptor imaging. (a) Five CHO cell lines transfected with different human RTKs were fixed, stained and imaged sequentially with 5 rounds of Exchange-PAINT (imager strands at 2.5 nM, 15000 frames, 10 Hz imaging rate). The diagonal images show the specificity of the imaging method and the specificity of the antibody-DNA conjugates for their cognate targets. Scale bars: 5 µm. (b) Specificity quantification of images shown in panel a. Localizations were normalized in each group by their maximal number and then the mean was calculated. There are significant differences in the number of localizations for each specifically transfected cell line. *P < 0.0001 ANOVA test, confirming high specificity.
