Oncogenic microRNA-181b is a negative regulator of ALFY mRNA in APL cell. (a) H1299 cells were seeded, transfected with a vector expressing a firefly luciferase under the control of the ALFY 3′UTR or together with a scrambled sequence (CTRL), miR-138 (138) or miR-181b (181). 24-hours later, luminescence was measured and the results are shown as n-fold regulation compared to the vector-only transfected cells. (bd) NB4 cells were transduced with lentiviral vectors to over-express miR-181b (181) or a scrambled sequence (CTRL). Total RNA was isolated at day 4 of ATRA treatment. Values were normalized for the expression of the SNORA66B and SNORA66 reference genes and are shown as n-fold regulation to untreated control transduced (CTRL) cells. In parallel, qPCR analyses of ALFY, CEBPE and CSF3R were performed. Standard deviations are shown for two independent experiments. Mann-Whitney-U test: *P < 0.05, **P ≤ 0.01.
