DNMT1-specific inhibition ameliorates the oscillatory shear (OS)-induced endothelial dysfunction. (a) ECs were exposed to pulsatile shear PS(12 ± 4 dynes/cm2) or OS (0.5 ± 4 dynes/cm2) for indicated time, and expression of DNMT1 was analyzed by Western blot. (b–e) ECs were infected with the control adenovirus (CL) or adenovirus expressing shRNA targeting human DNMT1 (shDNMT1). The infected ECs were exposed to PS or OS for 6 hours, cell migration was assessed by scratch wound healing assay (b), cell proliferation was indicated by immunofluorescent staining of the proliferative marker, Ki67 (c), ROS production was assessed (d), and monocyte adhesion assay was performed to exam the cellular inflammatory response.
