Workflow of the automated quantification of periodic structures exemplified with the MPS of a cultured hippocampal neuron imaged by STORM. (a) First, the regions containing neuronal material are identified using a suitable Gaussian filter and intensity threshold on the nanoscopy image (neuron discrimination). Then, the image is divided in subregions of predefined size (1 μm × 1 μm in this case) which are then catalogued as containing or not neuronal material. (b,c) The direction of the axon/dendrite \({\theta }_{N}\) is determined on each subregion. (d) The two-dimensional Pearson correlation coefficient \({R}_{P}\) is computed between each subregion and the reference pattern, within the area of neuronal material and for a range of directions \(\theta \) and phases \(\phi \) of the reference pattern. Here, the maximum value of \({R}_{P}(\phi )\) vs. \(\theta \) is shown for the subregions shown in (b) and (c).
