SDS-PAGE of heat-treated extracts from E. coli clones expressing EngU variants. M is the protein molecular weight marker, 10 µg protein per lane were loaded. The names of EngU derivatives are as in Fig. 2. The in silico predicted molecular weights are 100.2 kDa for the full-length EngU (containing parts A1BA2C, or amino acid positions 24 to 905), 46.9 kDa for A1B (containing amino acid positions 24 to 432 of EngU) and 54.5 kDa for A2C (containing amino acid positions 421 to 905 of EngU). The coding regions for parts written in square brackets were cloned in the first MCS, coding regions for C-terminally located enzyme parts were inserted into the second MCS of the pETDuet vector. This figure was assembled from two SDS PAGE gels.
