From: Deep mutational scanning of S. pyogenes Cas9 reveals important functional domains
Activity of additional positive selection enriched mutants. Ten mutations which were enriched after positive selection were tested for their ability to cleave chromosomally integrated GFP using eight different gRNAs. Their activity was normalized between WT SpCas9 activity with each gRNA and a non-targeting gRNA.
