Human CFSE+CD45+ 19F-PFC-labeled PBMC are present in the popliteal lymph node containing a positive 19F MRI signal. (a) Two days after the injection of 3 × 106 human 19F-PFC-labeled PBMC into the footpad of nu/nu mice, mice were subjected to 19F MRI and positive 19F MRI signal detection of 1.31 ± 0.2 × 105 cells (yellow arrow) resided within the popliteal lymph node (N = 3, n = 4/experiment; scale bar = 5 mm). (b) Following MRI, the lymph nodes were removed, cryopreserved, sectioned and fluorescent microscopy revealed the presence of CFSE+ 19F-PFC-labeled human PBMC (representative of n = 8 that did not undergo MRI). The sections were counterstained with DAPI to reveal nuclei (pseudocoloured red; 100x magnification, scale bar = 10 μm). (c) In a similar experiment, draining popliteal lymph nodes were removed and single cell suspensions prepared and counterstained with an antibody specific for human CD45. Flow cytometry analysis revealed the presence of CFSE+ human CD45+ 19F-PFC-labeled PBMC (c, representative N = 3, n = 4/experiment).
