IL-33 stimulated cytokine production is ST2-dependent and enhanced by IgE-mediated sensitisation of PDMCs and BMMCs. PDMCs (af) and BMMCs (g,h) derived from WT or ST2−/− (BALB/c) mice were sensitized (ah) or not (a–d) with murine anti-DNP IgE (0.5 μg/ml) overnight then stimulated with IL-33 (100 ng/ml, (ac); 10 ng/ml, e-h) for 24 h at 37 °C before release of IL-6, IL-13 or CCL2 was measured by ELISA. Data are presented as the mean values of triplicate samples ± SD and are from single experiments representative of 2 (ac) and 3 (eh) independent experiments. Statistical analysis was performed using One-way ANOVA with Bonferroni’s multiple comparison test where *p < 0.05 and **p < 0.01. Flow cytometric analysis (d) of PDMCs sensitized (black line), or not (red line), with murine anti-DNP IgE overnight were gated for live 7AAD- cells and then analysed for ST2 expression against the relevant isotype control obtained with non-sensitised cells (grey plot).
