Z-IETD-FMK and anti-FasL antibody partially inhibited Cd-induced apoptosis. (A) Primary rat cerebral cortical neurons and PC12 cells were pretreated with Z-IETD-FMK (40 µM) for 30 min prior to treatment with Cd (10 μM) for 24 h. After staining with annexin V-FITC, fluorescence microscopy analysis of neuronal cells was performed with a DMI3000 B inverted phase microscope at 200× magnifications using FITC and bright field filters. PC12 cells were pretreated with (B) Z-IETD-FMK (40 µM) or (C) anti-FasL (10 mg/L) for 30 min prior to treatment with Cd (10 μM) for 24 h. Following annexin V-FITC and PI double staining, the apoptosis rate of PC12 cells was analyzed by flow cytometry. A representative experimental result is shown. The apoptosis rate was calculated and analyzed using early apoptotic (Q4) and late apoptotic (Q2) cells. Data are expressed as mean ± SD (n = 3). **P < 0.01.
