Sensitivity analysis of real-time PCR assay for the identification of small hive beetle using plasmid DNA containing COI inserts. Plasmid containing COI insert of SHB were series diluted to create calibration curves for sensitivity calculations. The standard curve built from Cq values against the log copy number (range = 107–100 copies) of COI insert (n = 3). The 95% confidence intervals of the slopes were plotted with a blue line for VAITC5359 (Australia sample) and a red line for AQ4 (USA sample). The R2 = 0.993 for VAITC5359 and 0.998 for AQ4 were obtained for the assay.
