Effect of single point mutations on DPI-induced activation of GPR3. Dose-response curves for DPI in HEK293 cells expressing WT and single point mutants of GPR3. Twenty four hours after transfection, cells were stimulated for 30 minutes with increasing concentration of DPI and the intracellular cAMP level was measured. cAMP values were normalized to the maximal response. Nonlinear regression analysis was performed to generate dose-response curves and calculate concentration for 50% of the maximal effect (EC50). Data are the mean ± SD of 3 independent experiments.
