Glycan microarray analysis of anti-STn L2A5 mAb. The L2A5 hybridoma supernatant was diluted 1:2 and the results are shown as the means of fluorescence intensities of duplicate glycan probe spots, printed at 2 and 5 fmol (light and dark grey, respectively). The plot identifies the positive hits obtained. Imaging parameters are PMT 350, laser power 20%. In the inset, the laser scanning conditions were enhanced (PMT 350, laser power 90%) to highlight the binding of L2A5 to the α2–6-linked sialylated core-1 probes. The sequences of sialylated and mucin O-glycan core probes in the microarray are listed in Table 3.
