IRF1 knockdown decreases IFN-γ-induced PD-L1 expression. (A,B) Primary PSCs and the pancreatic stellate cell line were transfected with control siRNA or with siRNA against IRF1 for 48 hours. The cells were then treated with IFN-γ (0.2 μg/mL) for 24 hours. The effect on IRF1 was determined by qRT-PCR and by Western blotting, and the effect on PD-L1 mRNA was determined by qRT-PCR. IRF1 and GAPDH bands from three independent experiments were quantified by densitometry and data expressed as relative ratios.
