Monensin inhibits cell cycle progression and induces apoptosis in human pancreatic cancer cells. (A) Cell cycle analysis. Exponentially growing Panc-1 (a) and MiaPaCa-2 (b) cells were treated with monensin (2 µM) or vehicle control for 48 h. Cells were collected and stained, then subjected to FACS analysis. Percentages of cells in sub-G1 phase and S phase were graphed. “*” p < 0.05 and “**” p < 0.01, compared with that of the control groups. (B) Annexin-V apoptosis assay. Panc-1 (a) and MiaPaCa-2 (b) cells were treated with monensin (4 µM) or vehicle control. At 48 h post treatment, cells were collected and stained with Annexin V-FITC and propodium iodide, and subjected to flow cytometry. Average percentages of apoptotic cells (including early apoptosis and late apoptosis) were calculated and graphed. “**” p < 0.01, compared with that of the control groups.
