Verification of purity of the stria vascularis in the samples. (a) Cellular components of the lateral wall. The stria vascularis (StV) is composed of marginal, intermediate, and basal cells, whereas the spiral ligament (SL) consists of fibrocytes. This scheme was adapted from our earlier work57. (b) Assessment of purity of the StV in the obtained samples. qPCR analyses were performed separately on the StV (green bars) and SL (yellow bars) isolated from the lateral cochlear wall. In these assays, the primers for genes encoding K+,Cl-cotransporter type 3 (KCC3) and Brn-4, which are exclusively expressed in SL, and those for barttin and claudin 11, which are specific to StV, were used. Quantitative data (n = 3) were collected with reference to an internal gene, glyceraldehyde 3-phosphate dehydrogenase (GAPDH); averages, standard errors, and P values determined by two-tailed Student’s t test are also shown.
