Impaired immunosuppressive function of Tregs from nephritic PD-L1/ mice. (A) Tregs from nephritic PD-L1/ and WT mice were co-cultured with CD4+ CD25 T cells from CD45.1 mice in the presence of anti-CD3 antibody for three days. Responder CD4+ T-cell proliferation and activation were analyzed by flow cytometry. Representative dot plots are depicted. Mean ± SEM of one experiment out of three experiments are shown. (B) CD4+ CD25 T cells were adoptively transferred into Rag1/ mice either alone or together with Tregs from nephritic PD-L1/ or WT mice. NTN was induced two days later and mice were analyzed 8 days after NTN induction. Crescent formation was quantified in PAS-stained kidney sections. (C) Albumin-creatinine-ratio was determined in urine by ELISA. (D) Tubulointerstitial injury was quantified in PAS-stained kidney sections. Mean ± SEM of two experiments with 5 mice per group are shown. *p < 0.05; ns, not significant; w/o: without Tregs.
