The data were analyzed using Student’s t-test and 5–6 mice were used per genotype. The data are expressed as the mean ± SEM. (e) cathepsin-K-positive and (f) Mmp9-positive multinucleated osteoclast cells were quantified using IHC in the metaphyseal region of femurs from wild type and Sh3pxd2b-KO mice. Representative images of the stainings are shown. The number of osteoclasts/field in three wild type and three Sh3pxd2b-KO mice were analyzed, and the data are reported as the mean ± SEM. The scale bar represents 50 μm. The osteoclast marker RT-PCR measurements were performed separately in femur BM (g) and hard bone tissue (h). The expression levels of the TRAP and Mmp9 bone resorption markers were measured. Three 7-month-old mice were used per genotype. The data are presented as the mean ± SEM, and the differences were analyzed by Student’s t-test. *p < 0.05.
