Ca2+-binding energy and stoichiometry. (a) Isothermal Titration Calorimetry of Ca2+ binding to the D116A-MIIA domain. Data were acquired at 25 °C with 54 µM of protein and injection of 10 µL aliquots of 2 mM Ca2+ in the presence of 20 mM Tris-SO4, pH 7. Two binding sites were fitted with 54 and 57 kJ (±1.1 kJ, and χ2 /(degree of freedom) = 1.1). (b) Mass spectra representing the +7 charge states of the metal-free D116A-MIIA domain (green) and of the Ca2+- (blue) and Eu3+-(red) containing samples. Yellow diamonds indicate the shifted main peaks containing one or two bound metal ions.
