Rh2-induced membrane permeability is faster in the presence of eSM and the absence of Chol. LUVs composed of eSM:ePC (1:1, dashed lines), ePC:Chol (1:1, grey lines) or eSM:ePC:Chol (1:1:1, black lines) entrapping calcein at self-quenching concentrations were incubated with increasing Rh2/lipid ratios (indicated in the top right corner of each graph) at 25 °C. Membrane permeability was determined by the release of calcein from LUVs. The changes of fluorescence intensities were measured during 200 s, using λex and λem at 472 nm and 512 nm, respectively. Fluorescence intensities were normalized as described in Material and methods. Results are means of three independent experiments in triplicates.
