RN2N specificity and binding are retained in all antibody formats. (A) Gel electrophoresis and Coomassie staining of purified antibody formats shows that the 2N tau-specific RN2N antibody in an IgG format is approximately 156 kDa in size, the Fab is approximately 52 kDa in size and the scFv is approximately 29 kDa in size. (B) RN2N in all formats was shown to bind to full-length human tau using ELISA. (C) The single-cycle kinetics of RN2N to full-length human tau was determined using surface plasmon resonance. The KD of RN2N IgG was 381 nM, consistent with Fab (298 nM) and scFv (460 nM).
