(a) HEK293 cells were transfected with Pin1 plasmids (0.2, 0.5, 1, 2 μg per 100 mm dish), incubated in hypoxia for 8 h, and subjected to western blotting. (b) Cells were transfected with the indicated plasmids and siRNAs, incubated in hypoxia for 8 h, and subjected to western blotting. (c) Cells were transfected with the FLAG-fused 3′-UTR of PIN1 variant 1 or the FLAG-fused reverse sequence of the 3′-UTR plasmid. After cells were incubated in normoxia or hypoxia for 8 h, proteins were analyzed by western blotting. (d) Cells were transfected as indicated, incubated in hypoxia for 8 h, and subjected to western blotting. The expression of PIN1 3′-UTR was verified by examining FLAG expression.
