Inhibition of NFκB signalling reduces RV-induced expression of IFNβ and pattern recognition receptors in M1 differentiated THP-1 cells while increasing expression of IFNβ in M2 differentiated THP-1 cells. THP-1 cells were differentiated towards an M1 and M2 phenotype by stimulation with IFNγ and IL-4/IL-13, respectively. Macrophages were infected with RV16 for 24 h. At RV infection (+) or at the time of cytokine stimulation and after RV infection (++) the IκB kinase inhibitor PS1145 was added. Gene expression of IFNβ (A, B), RIG-I (C), MDA5 (D) and TLR3 (E) was measured by real-time PCR and data is presented as mean ± standard deviation fold change of untreated M1 differentiated macrophages relative to UBC/GAPDH expression (BE) or expressed as fold change of RV infected macrophages relative to UBC/GAPDH expression (A). Comparison of different groups was performed by one-way ANOVA with Tukey post- testing. *p < 0.05, **p < 0.01. Data from 4 independent experiments.
